[연구소식] 겐트대 생명의학연구센터 논문 발표, 기생충 DNA 진단 테스트 개발

DNA기반 새 진단 시험이 아시아의 병원성 기생충 퇴치에 희망을 가져오다.

 

Trypanosoma evansi (T. evansi) 라 불리는 기생충은 주로 농장 동물 및 야생 동물에 기생하는 것으로 알려졌으나, 최근 사람의 목숨까지 위태롭게 할 수 있다는 새로운 사실이 밝혀졌다. 트리파노소마 기생충은 아프리카에서 발견되었지만 운송된 감염 동물에 의해서 아시아로 확산되었다. 흡혈 곤충과 박쥐에 의해서 병균이 전달되기 때문에 새로운 진단 도구 없이는 병균의 확산을 막기 힘들다.

 

최근 아시아에서 늘어난 감염병 관련 사건들를 연구를 위해 겐트대학교 글로벌캠퍼스의 생명의학연구센터 (Radwanska 교수, Magez 교수와 방문학생 Julie Goossens)와 중국 항저우에 위치한 저장의학과학원(Shaohong Lu 교수, 浙江医学科学院)의 공동연구프로젝트가 추진 되었다. 이 공동연구는 중국과 벨기에 국가연구펀드로 지원되었고 2017년에 체결한 MOU를 통해 진행되었다. 최근에는, 이번 협업을 통해 치명적인 기생충 감염을 효과적으로 탐지하고 치료할 수 있는 빠른 진단 테스트가 개발되었다.

 

이 실험은 LAMP라는 매우 특유한 DNA 증폭 테스트에 기반을 둔다. LAMP 테스트는 T. evansi 감염으로 인해 가축 사육과 인간에 위협이 될 수 있는 지역에서 기생충 방제 및 제거를 위한 전략적인 도구로 사용될 수 있다. 이 테스트의 새로운 점은 샘플을 전문 실험실에 가져올 필요없이 현장 조건에서 매우 간단한 도구와 장비를 사용하여 DNA 검출을 수행 할 수 있는 것이다. 따라서 시험은 농장 주인의 참석 하에 수행 할 수 있으며 감염된 동물은 즉시 치료될 수 있다.

 

이번 공동연구의 결과는 Elsevier의 수의기생충저널 (Veterinary Parasitology) 2018년 1월호에 발표되었으며 2018년 5월에 열리는 ‘제10회 수의학 및 동물 과학 세계대회’에서 발표 될 예정이다. 이 연구는 또한 2017년 12월 15일, Magez 교수 지도하에 박사 학위를 취득한 브뤼셀 자유대학교 (Vrije University in Brussels) Julie Goossens 박사의 논문과제이다.

 

Lu교수와 Kang교수는 2018년 1월 21일 GUGC 캠퍼스를 방문하여 GUGC-BMRC 그룹과 2일 간의 공동 워크샵을 진행했다. 이번 워크샵을 통해 새로운 공동 연구 프로젝트를 추진하였고 또한 저장대학에서 한 명의 학생이 GUGC에서 와서 4년 박사 학위과정을 진행할 수 있게 되었다. 저장대학교는 상하이 순위에 따라 중국 3위의 대학이며 GUGC는 중국 파트너와의 긴밀한 협력을 계속 진행 할 수 있는 토대를 구축하였다.

원문: New DNA based diagnostic test helps to eradicate pathogenic parasite in Asia

 

Trypanosoma evansi (T. evansi) is a parasite that infects mainly commercial farm animals and wildlife. In recent years however it was reported that the parasite can also infect and kill humans. In the past, most trypanosome parasites were found in Africa, but T. evansi has been imported into Asia by the accidental shipping of infected animals. As the parasite is transmitted by biting and blood sucking insects, and even vampire bats, it is very hard to stop the spread of the disease without good diagnostic tools. A recent increase in numbers of cases occurring in Asia prompted a collaborative research projected between the Biomedical Research Center (BMRC) at the Ghent University Global Campus, South Korea (Prof. Magdalena Radwanska & Prof. Stefan Magez and Julie Goossens, a visiting student from the Magez laboratory at the Vrije University in Brussels) and the Zhejiang Academy of Medical Sciences, Hangzhou, China (Prof. Shaohong Lu). This collaboration has been supported by national grants from both China and Belgium (FWO-G013518N), and has been enforced by a 2017 MOU between the partners. Most recently, the collaboration resulted in the development of a rapid diagnostic test allowing for effective detection and treatment of this deadly parasite infection. The test is based on a highly specific DNA amplification test called LAMP. The LAMP test could be used as a tool for parasite control and elimination strategies in areas where T. evansi infections are causing a threat to livestock farming and where potential transmission to humans occurs. The novelty of the test is that although it involves DNA amplification, it uses very simple tools and equipment that allow for the first time to perform DNA detection in field conditions, without having to bring the samples to a specialized laboratory. Therefore the test can be done in the presence of the animal owner, and infected animals can be treated immediately. The results of this collaborative work have been just published in January’s 2018 issue of the Veterinary Parasitology, and the work will be presented at the 10th World Congress on Veterinary and Animal Science in May, in Osaka, Japan. The work was also part of the PhD of Julie Goossens who defended her PhD under the promotorship of Prof. Magez on December 15th 2017.

 

Prof. Lu accompanied by Dr. Kang visited the GUGC campus on January 21st for a joint 2-day workshop with the GUGC-BMRC group. A new collaborative PhD project was initiated during this meeting and will allow a student from the Zhejiang University to conduct his Ph.D at GUGC during the next 4 years. Zhejiang University is the 3rd ranking Chinese University according to the Shanghai ranking and we are excited to have a continued close collaboration with our Chinese partners.
