겐트대 박지혜 박사, 한국연구재단 과제 수주

 

5월25일, 겐트대학교 글로벌캠퍼스 식물바이오활성물질연구센터 (GUGC Plant Bioactive Compound Research Centre) 의 Postdoc 과정을 밟고 있는 박지혜박사가 한국연구재단에 신청한 연구과제가 ‘2018년 이공학개인기초연구지원사업’에 선정되었다. 이로서 GUGC 사상 최초로 개인연구자가 NRF과제수주에 성공한 사례가 탄생되였다. 한국연구재단 이공학개인기초연구지원사업의 목적은 이공학분야 풀뿌리 개인기초연구를 폭넓게 지원하여 변혁적 연구기반을 확대하고 국가 연구역량을 제고하며, 연구자가 안정적으로 연구에 몰입할 수 있는 환경을 조성하여 창의적 연구를 활성화시키는 것이다. 2018년도 최종 선정 현황으로는 기본 연구 2,408건, 보호 연구 7건, 지역대학우수과학자 238건, 한국형SGER 98건, 기본 연구(후속) 7건, 및 지역대학우수과학자(후속) 19건이다.

 

이번 한국연구재단에 채택된 과제 제목은 ‘갯벌 저서성 기형 및 지질체 형성 규조류의 분자화학생물다양성과 생리생태학적 기초 연구 (A basic study on molecular chemobiodiversity and ecophysiology of periphytic diatoms (showing deformity and lipid body formation) collected from a tidal-flat ecosystem) ’ 이며, 박지혜 박사는 이 연구과제를 통해, 외국대학이 국내에서 국가과제를 신청하는 데 겪고 있는 어려운 상황에서 본인의 과제가 GUGC 연구에 기여가 되기를 바란다는 소감을 밝혔다.

 

박지혜 박사는 2017년 2월에 겐트대학교 글로벌캠퍼스와 인천대에서 첫 번째로 공동 박사학위를 받았으며, 주 연구영역은 분자식물학으로 해조류 세포 및 분자매커니즘에 대한 연구를 통해 해양환경보호와 바이오소재의 지속 가능한 응용이다. 박지혜 박사의 파래에 대한 연구결과는 최근에 Elsevier 출판의 북 챕터에 저술되었다.

 

 

 

박지혜 박사의 최근 발표된 북 챕터 개요:

구멍갈파래 (U. purtusa)는 지중해, 태평양 및 인도양에 널리 분포하는 갈파래류 (Ulva spp.)에 속하는 녹조류이다. 최근, 산업활동 및 도심으로부터 유래하는 유독성 폐기물이 빈번하게 연안 해역에 침범하는데, 갈파래는 이를 탐지할 수 있는 중요한 지표종으로 보고된 바 있다. 갈파래는 연안 생태계의 기초를 구성하는 일차 생산자로서 다른 해양 생물에게 서식지를 제공하고, 식품 및 의약품의 중요한 천연 자원으로 활용될 수 있다. 생식은 개체군의 유지에 필수적인 과정으로서 생식 과정에 문제가 생기면 이는 곧 개체군의 사멸을 의미하고 연쇄적으로 생물군집 전체에 변화를 초래하게 된다. 많은 갈파래는 지리적으로 넓은 범위로 분포하고 있으며 유사한 형태의 생식 과정을 가진다. 따라서, 본 표준 시험 방법은 전 세계 대부분 연안 해역에서 보고된 동일한 속(genus)에 속하는 다른 종(species)에 적용될 가능성이 있다. 여기서 제안하는 파래를 이용한 생태독성 기법은 종래에 사용되는 기술에 비해 많은 실질적인 이점을 제공한다. 몇 가지 예를 들자면, 전문적 지식이 요구되지 않는다, 96시간 노출시간에 시험 분석은 3시간 정도 소요되며, 필요한 시료의 양은 매우 적고 시험 용기도 웰 플레이트만 요구되기 때문에 비용적으로도 효율적인 방법이라고 할 수 있다. 구멍갈파래 시험법의 민감도는 일반적으로 이용 가능한 또는 잘 확립된 생물검정방법과 유사하며, 많은 경우에 있어서 기존의 방법보다 더욱 우수하며, 생식은 생물의 번식이 촉진되는 수단이므로 생태학적으로 매우 중요한 종말점이라고 할 수 있다. 또한, 야외에서 채집된 갈파래는 쉽게 실내 배양을 유지할 수 있고 실험실에서 인공 생식 유도를 할 수 있다. 구멍갈파래 시험방법은 연안 및 기수역 생태계를 모니터링하기 위해 기존에 사용되는 방법에 추가적으로 적용할 수 있는 가치 있는 방법이 될 수 있다. 본 시험방법은 폐수에 대한 TIE/TRE 절차에도 적용될 수 있다. 이 기술은 기존에 제안한 ISO 방법을 기반으로 개발되었기 때문에 폐수 관리 기술에도 쉽게 응용되어 개발될 수 있으며, 보다 간편한 사용을 위해 Ulva키트와 자동화기기인 UlvaTOX도 개발된 바 있다.

 

Ulva pertusa Kjellman is reported from the Mediterranean, Pacific, and Indian Oceans. Ulva spp. are important representatives of macroalgal communities in coastal waters that are under threat from the frequent inundations of toxic waste derived from industrial and municipal sources. These algae are primary producers constituting the basis of costal ecosystem and, moreover, provide shelter and habitats for other marine organisms; they are also important natural sources of food and pharmaceutical products. Reproduction is a critical process by which populations perpetuate, and disturbance of this process can cause failure in recruitment, leading to the disappearance of the population and ultimately to modifications in community structure and dynamics. Members of Ulva have a wide range of geographic distribution and undergo similar reproductive processes. Therefore, the test method presented in this standard has the potential for application to other species belonging to the same genus reported from coastal waters in most parts of the world.

The proposed macroalgal toxicity test provides many practical advantages over other currently employed techniques. No specialist expertise is required. The test is cost- and time effective, since it only requires a cell plate, a small volume of water and takes a total of approximately 3 h to conduct around a 96 h incubation period. The sensitivity of the Ulva method is similar to, and in many cases better than, commonly available or well established bioassay methods, and since reproduction is the means by which population recruitment is facilitated, the measured endpoint being ecologically significant cannot be disputed. In addition, field-collected samples of unialgal Ulva plants can easily be held and acclimated in a holding tank for 1 to 2 month(s), and artificial induction of reproduction in a laboratory is easily achieved using vegetative thalli, allowing all year round testing to be performed. The Ulva reproduction bioassay would be a valuable addition to the existing suite of tests routinely used to monitor coastal and estuarine ecosystems.

The Ulva test can also be applicable to the TIE/TRE procedures for waste waters. The TIE/TRE technique using Ulva may signify a break-through in waste-water management technology since no such technique has been developed so far based on the established ISO methods.

The Ulva-Kit and automated UlvaTOX have been developed to facilitate the use of the Ulva method.
